چکیده
AIM To determine whether Lactobacillus plantarum can modify the deleterious effects of tumor necrosis factor-alpha (TNF-alpha) on intestinal epithelial cells. METHODS Caco-2 cells were incubated with TNF-alpha alone or in the presence of L. plantarum. Transepithelial electrical resistance was used to measure epithelial barrier function. Interleukin 8 (IL-8) secretion by intestinal epithelial cells was measured using an ELISA. Cellular lysate proteins were immunoblotted using the anti-extracellular regulated kinase (ERK), anti-phospho-ERK and anti-IkappaB-alpha. RESULTS A TNF-alpha-induced decrease in transepithelial electrical resistance was inhibited by L. plantarum. TNF-alpha-induced IL-8 secretion was reduced by L. plantarum. L. plantarum inhibited the activation of ERK and the degradation of IkappaB-alpha in TNF-alpha-treated Caco-2 cells. CONCLUSION Induction of epithelial barrier dysfunction and IL-8 secretion by TNF-alpha is inhibited by L. plantarum. Probiotics may preserve epithelial barrier function and inhibit the inflammatory response by altering the signal transduction pathway.

Bacteria in the vaginal microbiome alter the innate immune response and barrier properties of the human vaginal epithelia in a species-specific manner.
BACKGROUND Bacterial vaginosis increases the susceptibility to sexually transmitted infections and negatively affects women's reproductive health. METHODS To investigate host-vaginal microbiota interactions and the impact on immune barrier function, we colonized 3-dimensional (3-D) human vaginal epithelial cells with 2 predominant species of vaginal microbiota (Lactobacillus iners and Lactoba...
متن کاملLysate of Probiotic Lactobacillus plantarum K8 Modulate the Mucosal Inflammatory System in Dextran Sulfate Sodium-induced Colitic Rats
Inflammatory bowel disease (IBD) is caused by dysregulation of colon mucosal immunity and mucosal epithelial barrier function. Recent studies have reported that lipoteichoic acid (LTA) from Lactobacillus plantarum K8 reduces excessive production of pro-inflammatory cytokine. In this study, we investigated the preventive effects of lysate of Lb. plantarum K8 in dextran sulfate sodium (DSS)-induc...
متن کاملTumor necrosis factor-alpha regulates inflammatory and mesenchymal responses via mitogen-activated protein kinase kinase, p38, and nuclear factor kappaB in human endometriotic epithelial cells.
Tumor necrosis factor (TNF)-alpha is central to the endometriotic disease process. TNF-alpha receptor signaling regulates epithelial cell secretion of inflammation and invasion mediators. Because epithelial cells are a disease-inducing component of the endometriotic lesion, we explored the response of 12Z immortalized human epithelial endometriotic cells to TNF-alpha. This report reveals the im...
متن کاملKK/Ta Mice Administered Lactobacillus plantarum Strain No. 14 Have Lower Adiposity and Higher Insulin Sensitivity
Excess accumulation of white adipose tissue can lead to obesity-related metabolic abnormalities such as insulin resistance. We previously reported that intragastric administration of Lactobacillus plantarum No. 14 reduced adipocyte size in diet-induced obese C57BL/6 mice. The present study tested whether L. plantarum No. 14 affects adiposity and insulin sensitivity in an animal model of type-2 ...
متن کاملInduction of interleukin-8 secretion and apoptosis in bronchiolar epithelial cells by Fas ligation.
Epithelial cell injury is the common manifestation of lung injury. Contributing to such injury of epithelial cells is apoptosis. Although apoptosis is part of the normal process of epithelial renewal, in excess it is pathologic. We previously demonstrated the excessive apoptosis of lung epithelial cells and the upregulation of Fas and Fas ligand (FasL) in fibrosing lung diseases. We also showed...
